The cysteine-rich secretory proteins (CRISP) family is a group of four proteins that are strongly expressed in the male reproductive tract and have been implicated in having roles in male fertility. CRISP2, also known as TPX1, has been implicated in the adhesion between spermatids and Sertoli cells, and with CRISP1, is thought to be involved in sperm-egg fusion. CRISP2 has been shown to regulate the Ca2+ influx through ryanodine receptors (RYR) and may influence the acrosome reaction or sperm motility. CRISP2 has also been shown to bind to the mitogen-activated protein kinase kinase kinase 11 (MAP3K11) and localizes to the developing acrosome, suggesting this CRISP2-MAP3K11 complex may have a role in acrosome development.

FAM120A (C9orf10) is a member of the constitutive coactivator of PPAR gamma family and the gene was mapped to chromosome 9q22.31. FAM120A was recently detected within the Pur-alpha-containing mRNA-protein complex in the brain. As a novel RNA-binding protein, FAM120A is a critical component of the oxidative stress-induced survival signaling. It may participate in mRNA transport in the cytoplasm. FAM120A activates src family kinases and acts as a scaffolding protein enabling src family kinases to phosphorylate and activate PI3-kinase. FAM120A protects cells from apoptosis through activation of SRKs in response to oxidative stress. Blocking of the survival signaling mediated by FAM120A, which sensitizes the cancer cells to stress-induced apoptosis, may be a novel therapeutic approach for gastric scirrhous carcinoma cells.

Calpains make up a ubiquitously expressed, well-conserved family of calcium-dependent cysteine proteases. The calpain proteins are heterodimers consisting of an invariant small subunit and variable large subunits. This large subunit possesses a cysteine protease domain, and both subunits possess calcium-binding domains. Calpains have been implicated in neurodegenerative processes as their activation can be triggered by calcium influx and oxidative stress. Calpain 6 (CAPN6) is most similar to Calpain 5; the C-terminal region of CAPN6 lacks homology to the calmodulin-like domain of other vertebrate calpains. CAPN6 is thought to be involved in the regulation of microtubule dynamics and cytoskeletal organization. CAPN6 has also been recently identified as an HIV dependency factor (HDF), suggesting that CAPN6 may be an important drug target in HIV treatment.

Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand of the TNF family through their death domain containing receptors, TNFR1 and Fas. A novel cell death receptor was recently identified by several groups independently and designated DR3, Wsl-1, Apo-3, TRAMP and LARD1-5. The ligand for this novel cell death receptor has not yet been defined. DR3 is highly expressed in the tissues enriched in lymphocytes including PBL, thymus and spleen. Like TNFR1, DR3 induces apoptosis and NF-kB activation.

The coiled-coil domain is a common protein motif that is often involved in protein oligomerization and is found in proteins such as transcription factors and intermediate filaments. One such protein is CCDC134, a recently identified secretory protein that has been found to inhibit the transcriptional activity of the Elk1 protein. Overexpression CCDC134 also inhibited the phosphorylation of Erk and JNK/SAPK but not p38 MAPK, while specific siRNA against CCDC134 activated Elk1 transcriptional activity and the phosphorylation of Erk and JNK/SAPK, suggesting a potential inhibiting role of CCDC134 in MAPK-mediated Elk1 transcription. CCDC134 is widely expressing in normal adult tissues, tumors, and cell lines.

BRSK1 was initially identified as a mammalian homolog to the fission yeast S. pombe Cdr2, a mitosis-regulatory kinase and also shows significant homology to the C. elegans neuronal cell polarity regulator SAD1. BRSK1 is unbiquitously expressed, with highest levels of expression in the brain and testes. Similar to its yeast homolog, BRSK1 is thought to be involved in stress-induced cell cycle arrest. Overexpression of this protein leads to the G2/M arrest in HeLa S2 cells and UV-induced G2/M arrest could be partially abrogated by reduced expression of BRSK1 through the use of siRNA, indicating its role in DNA damage checkpoint function. More recently, it has been shown that both BRSK1 and the related protein BRSK2 are required for mammalian neuronal polarization. While BRSK1- and BRSK2-null mice were viable, double-mutant mice died within two hours of birth. Neurons from these mice showed uniformly-sized neurites as opposed to the normal long axon and multiple shorter dendrites. These neurites also displayed both axonal and dendritic markers. At least two isoforms of BRSK1 are known to exist.

Cell cycle regulated protein ubiquitination and degradation within subcellular domains is thought to be essential for the normal progression of mitosis. APC13 is a highly conserved component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC/C is responsible for degrading anaphase inhibitors, mitotic cyclins, and spindle-associated proteins ensuring that events of mitosis take place in proper sequence. The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. These death signals finally cause the degradation of chromosomal DNA by activated DNase. A human 45 kDa DNA fragmentation factor (DFF45) was identified recently which was cleaved by caspase-3 during apoptosis. Mouse homologue of human DFF45 was identified as a DNase inhibitor designated ICAD. Upon cleavage of DFF45/ICAD, a caspase activated deoxyribonuclease (DFF40/CAD) is released and activated and eventually causes the degradation of DNA in the nuclei. Therefore, the cleavage of DFF45/ICAD, which causes DFF40/CAD activation and DNA degradation, is the hallmark of apoptotic cell death.

EBI3 is a subunit in two distinct heterodimeric cytokines: interleukin-27 (IL-27) and IL-35. Like interleukin-23 (IL-23), IL-27 is a recently discovered member of the IL-6/IL-12 family of proinflammatory and immunoregulatory cytokines. It exists as a heterodimer composed of the p40-related protein EBI3 and an IL-12 p35-related protein termed p28. IL-27 is produced after activation by antigen-presenting cells and induces proliferation of naïve but not memory CD4+ T-cells. It acts by binding to its receptor WSX-1 (also known as TCCR) and gp130 which results in the activation of a Jak/STAT signaling cascade, suggesting the IL-27 is involved in the regulation of immune processes. It has been suggested that IL-27 can also be used as a therapeutic agent against cancer as it can also induce tumor-specific anti-tumor activity mediated through CD8+ T-cells, IFN-gamma, and T-bet. IL-35 is composed of EBI3 and the p35 subunit of IL-12 and has been reported to have therapeutic effects against collagen-induced arthritis by expanding the population of regulatory T cells and suppressing Th17 cells.
