DEAD box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp, are putative RNA helicases implicated in several cellular processes involving modifications of RNA secondary structure. DHX36 (DEAH box protein 36), also known as MLE-like protein 1 and RNA helicase associated with AU-rich element ARE (RHAU), belongs to RNA helicase of the DEAH family and may function in sex development and spermatogenesis. It is expressed in testis and is evolutionary conserved with true orthologs in almost all animal species. DHX36 plays a role in degradation and deadenylation of mRNAs containing in their 3'-UTR the consensus ARE sequence element. DHX36 is required for early embryogenesis.

Adipose tissue of an organism plays a major role in regulating physiologic and pathologic processes such as metabolism and immunity by producing and secreting a variety of bioactive molecules termed adipokines. One highly conserved family of adipokines is adiponectin/ACRP30 and its structural and functional paralogs, the C1q/tumor necrosis factor-alpha-related proteins (CTRPs) 1-7. Unlike adiponectin, which is expressed exclusively by differentiated adipocytes, the CTRPs are expressed in a wide variety of tissues. These proteins are thought to act mainly on liver and muscle tissue to control glucose and lipid metabolism. An analysis of the crystal structure of adiponectin revealed a structural and evolutionary link between TNF and C1q-containing proteins, suggesting that these proteins arose from a common ancestral innate immunity gene. CTRP6 contains at least 4 glycosylation motifs, suggesting that CTRP6 may be highly post-translationally modified.

Fas-associated protein 1 (FAF1) was initially identified as a Fas-binding pro-apoptotic protein that is component of the death-inducing signaling complex in Fas-mediated apoptosis. FAF1 can also induce apoptosis in the absence of extrinsic death signals when overexpressed although it does not contain typical death motifs such as the death domain, death effector domain, and caspase recruitment domain. Overexpression of FAF1 also decreases the basal level of NF-kB activity in transfected 293 cells, inhibits NF-kB activity induced by TNF-a, IL-1b and lipopolysaccharide, and prevents NF-kB translocation to the nucleus, suggesting that another role of FAF1 is to negatively regulate the activity of NF-kB. FAF1 can also interact with the inflammatory signaling PYRIN-containing Apaf-1-like proteins (PYPAFs, also called NALPs) such as PYPAF1, PYPAF2 (NALP2), and PYPAF7, suggesting FAF1 may also be involved in the inflammation pathway. Collapsin-response mediator proteins (CRMPs) are highly expressed in the developing brain where they play major roles in axonal outgrowth, neurite differentiation, and apoptosis. Their continued expression in areas of high synaptic remodeling such as the cerebellum, hippocampus, and the olfactory system suggests that these proteins may also be involved in adult brain plasticity. CRMP-1 was initially identified as a dihydro-pyrimidinase expressed exclusively in brain; later studies have shown that it is involved with neurotrophin (NT) 3-induced neurite formation and outgrowth. CRMP-1 localization switches from axonal to somatodendritic when neurons reach functional maturity, suggesting that it is involved in early neuronal differentiation as well as in later processes related to the survival or death of the newly generated neurons.

CCR8 is one of the chemokine receptors that are required as coreceptors for HIV infection. The genes encoding human and murine CCR8 were cloned and designated TER1, CKR-L1, and ChemR1. The encoded seven transmembrane protein was identified as the receptor for human CC chemokine I-309 and renamed CCR8. Recently, CCR8 was found to serve as a coreceptor for diverse T-cell tropic, dual-tropic and macrophage-tropic HIV-1 strains. CCR8 mediates CC chemokine I-309 induced monocyte chemoattraction and HIV-1 envelope fusion and virus infection, which can be prevented by the CCR8 ligand I-309. CCR8 is expressed in spleen, thymus and T lymphoblastic cell lines.

Caspases are a family of cysteine proteases that can be divided into the apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens. The apoptotic subfamily can be further divided into initiator caspases, which are activated in response to death signals, and executioner caspases, which are activated by the initiator caspases and are responsible for cleavage of cellular substrates that ultimately lead to cell death. Caspase-8 is an initiator caspase that was identified as a member of the Fas/APO-1 death-inducing signaling complex. The adaptor molecule FADD couples procaspase-8 to the Fas receptor death domain; subsequent oligomerization promotes procaspase-8 autoactivation. FLIP, a catalytically inactive caspase-8-like molecule inhibits these interactions and thus can inhibit apoptosis. This antibody will only detect isoform E of caspase-8.

Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand of the TNF family through their death domain containing receptors, TNFR1 and Fas. A novel cell death receptor was recently identified by several groups independently and designated DR3, Wsl-1, Apo-3, TRAMP and LARD1-5. The ligand for this novel death receptor has been defined as TWEAK, also termed Apo3L. DR3 is highly expressed in the tissues enriched in lymphocytes including PBL, thymus and spleen. Like TNFR1, DR3 induces apoptosis and NF-kB activation.
