The FCH and double SH3 domains protein 1 (FCHSD1, also known as NWK2) and the related protein FCHSD2 were initially identified in silico as distantly related proteins to FNBP1 and FNBP2. Both share the common domain structure consisting of FCH, FBH, two SH3 and C-terminal proline-rich domains. While little is known of the function of FCHSD1, the related protein NWK is an adaptor protein that is thought to regulate Rho activity downstream of Robo receptors, suggesting that FCHSD1 may be involved in synaptic morphology by regulating actin dynamics in presynaptic terminals.

Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. The mammalian homologues of the key cell death gene CED-4 in C. elegans has been identified recently from human and mouse and designated Apaf1 (for apoptosis protease-activating factor 1). Apaf1 binds to cytochrome c (Apaf-2) and caspase-9 (Apaf-3), which leads to caspase-9 activation. Activated caspase-9 in turn cleaves and activates caspase-3 that is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis. Apaf1 can also associate with caspase-4 and caspase-8. Apaf1 is ubiquitously expressed in human tissues.

The susceptibility of primary splenic B cells to Fas-mediated apoptosis is regulated in a receptor-specific fashion. Terminal effectors of B cell Fas-resistance include the known anti-apoptotic proteins Bcl-xL, FLIP, and a recently identified protein termed FAIM. This molecule is broadly expressed in various tissues and exists in at least three isoforms. It is thought that resistance to Fas killing via increased expression of FAIM protects foreign antigen-specific B cells during interactions with FasL-bearing T cells whereas autoreactive B cells are deleted via Fas-dependent cytotoxicity. More recent results have indicated that FAIM interacts with both Trk and p75 neurotrophin receptor and may play a role in promoting neurite outgrowth in different neuronal systems by a mechanism involving the activation of NF-kB and the Ras-ERK pathway.

Caspases are a family of cysteine proteases that can be divided into the apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens. The apoptotic subfamily can be further divided into initiator caspases, which are activated in response to death signals, and executioner caspases, which are activated by the initiator caspases and are responsible for cleavage of cellular substrates that ultimately lead to cell death. Caspase-3 is synthesized as an inactive proenzyme that undergoes proteolytic cleavage by caspases 8, 9 and 10 to produce 2 subunits, termed p20 and p11. Mitogen-activated protein (MAP) kinase cascades are activated in response to various extracellular stimuli, including cytokines, growth factors and environmental stresses. A novel MAP kinase kinase kinase (MAPKKK) was recently identified and designated ASK1 (for apoptosis signal-regulating kinase 1) and MAPKKK5. ASK1 activated two different subgroups of MAPKK, MKK4 and MKK6, which in turn activated c-Jun N-terminal kinase (JNK) and p38 MAP kinase, respectively. ASK1/MAPKKK5 is activated by TNFR and Fas through the interaction with members of the TRAF family and Fas-associated protein Daxx. Rockland produces highly active antibodies and conjugates to collagens. Collagens are highly conserved throughout evolution and are characterized by an uninterrupted ''Glycine-X-Y'' triplet repeat that is a necessary part of the triple helical structure. For these reasons, it is often extremely difficult to generate antibodies with specificities to collagens. The development of ‘type’ specific antibodies is dependent on NON-DENATURED three-dimensional epitopes. Rockland extensively purifies collagens for immunization from human and bovine placenta and cartilage by limited pepsin digestion and selective salt precipitation. This preparation results in a native conformation of the protein. Antibodies are isolated from rabbit antiserum and are extensively cross-adsorbed by immunoaffinity purification to produce 'type' specific antibodies. Greatly diminished reactivity and selectivity of these antibodies will result if denaturing and reducing conditions are used for SDS-PAGE and immunoblotting.

CLEC2A (C-type lectin domain family 2 member A), also known as PILAR (proliferation-induced lymphocyte-associated receptor) is involved in modulating T-cell expansion. It is a 32 kDa single-pass type II membrane protein that contains one C-type lectin domain in its extracellular region and belongs to the CTL/CTLD superfamily. CLEC2A is mainly expressed in skin and highly expressed in CD8(+), B lymphocytes and naive CD4(+) T cells. Manipulation of CLEC2A signaling may be important for treatment of autoimmune diseases and cancer.
