CENPW was initially identified as a gene that was upregulated in multiple cancers, and whose overexpression in mouse fibroblast cells gave rise to distinct cancer-specific phenotypes. It was later found to be a nuclear protein that associated with CENPT, a component of CENPA nucleosome complex in the centromere, and is required for proper kinetochore function. CENPW also specifically interacts with the nucleolar phosphoprotein nucleophosmin, also known as B23 It has been suggested that nucleophosmin functions in the assembly of the kinetochore by interacting with CENPW during interphase. Overexpression of CENPW in the SKOV-3 human ovarian cancer cell line as well as in the zebrafish embryo led to apoptosis, suggesting that high levels of CENPW induces apoptotic cell death.

Apoptosis, or programmed cell death, is related to many diseases, such as cancer. Apoptosis is triggered by a variety of stimuli including members in the TNF family and can be prevented by the inhibitor of apoptosis (IAP) proteins. IAP proteins form a conserved gene family that binds to and inhibits cell death proteases. The two isoforms of c-IAP (c-IAP1 and c-IAP2) are structurally related to XIAP, containing 3 baculoviral IAP repeat (BIR) motifs that are essential and sufficient for the binding and inhibition of caspases-3, -7. The c-IAPs can associate with the death receptor TNF-R2, and mediate the ubiquitinization of TRAF2 following the binding of TNF-a by its receptor. Omi, a negative regulator of c-IAP, inhibits its activity by catalytically cleaving c-IAP. Another negative regulator, Smac/DIABLO, acts by enhancing the auto-ubiquitization activity of c-IAP.

TRAIL/Apo2L is a new member of the TNF family and induces apoptosis of a variety of tumor cell lines. DR4 and DR5 are the recently identified functional receptors for TRAIL. Two decoy receptors for TRAIL have been identified and designated DcR1/TRID/TRAIL-R3/LIT and DcR2/TRAIL-R4/TRUNDD. DcR1 has extracellular TRAIL-binding domain but lacks intracellular signaling domain. It is a glycophospholipid-anchored cell surface protein. DcR1 transcripts were expressed in many normal human tissues but not in most cancer cell lines. Overexpression of DcR1 did not induce apoptosis, but attenuated TRAIL-induced apoptosis.

CKIP1 was identified through a yeast-two hybrid screening as a protein that would interact with only one of the two catalytic subunits of the casein kinase 2 complex. CKIP1 is a pleckstrin homology domain-containing protein localized within the nucleus and at the plasma membrane that interacts with CK2alpha but not CK2alpha', and is thought to play a role in targeting CK2alpha to a particular cellular location. CKIP1 has been implicated in muscle differentiation and the regulation of cell morphology and actin cytoskeleton. CKIP1 can also interact with other proteins such as ATM, an upstream kinase of p53, and recruit the nuclear ATM to the plasma membrane, suggesting CKIP1 may regulate ATM function through re-localizing ATM. CKIP1 can also form a complex with the kinase AKT, leading to a down-regulation of PI3K/AKT signaling and suppression of tumor growth in vivo.

Mouse homologue of human DFF45 was identified as a DNase inhibitor designated ICAD. DFF45/ICAD have short forms that were termed DFF35 and ICADs, respectively. Upon cleavage of DFF45/ICAD, the caspase activated deoxyribonuclease (DFF40/CAD) is released and activated and eventually causes the degradation of DNA in the nuclei. Therefore, the cleavage of DFF45/ICAD, which causes DFF40/CAD activation and DNA degradation, is the hallmark of apoptotic cell death.

One downstream target of TOR is the eukaryotic initiation factor 4E binding protein 1 (4E-BP1) whose phosphorylation prevents its association with eIF4E, preferentially stimulating translation of mRNAs containing long, highly structured 5'-UTRs. Rapamycin inhibits TOR resulting in reduced cell growth and reduced rates of cell cycle and cell proliferationl, at least in part by inhibiting the activity of TOR towards 4E-BP1.
